چکیده

Uromodulin is well-known to be the more potent immunosuppressive glycoprotein isolated from pregnant women urine, compared to its counterpart, Tamm-Horsfall glycoprotein (THP) isolated from non-pregnant women/male urine. However, structural profiles of these glycoproteins were controversial until today. Hence, this paper aims to identify the pregnancy associated glycosylation changes of THP and uromodulin. THP and uromodulin isolated with diatomaceous earth filtration were subjected for structural characterization using MALDI-TOF tandem mass spectrometry (MS) analyses. Both THP and uromodulin expressed high-mannose and complex-type N-glycan carrying Sda, LacNAc, LacdiNAc sequence as the capping antennae. In comparison to THP, uromodulin expressed higher abundance of LacdiNAc sequence and heavily sialylated complex type Nglycans, where up to four sialic acid residues were observed in its tetra-antennary glycans. Both THP and uromodulin expressed Core 1 and Core 2 O-glycans. These findings suggested that the higher level of LacdiNAc sequence and sialylation in uromodulin might facilitate its lymphocyte suppressive response more effectively than that of THP.

Role of Tamm-Horsfall protein and uromodulin in calcium oxalate crystallization.

One of the defenses against nephrolithiasis is provided by macromolecules that modulate the nucleation, growth, aggregation and retention of crystals in the kidneys. The aim of the present study was to determine the behavior of two of these proteins, Tamm-Horsfall and uromodulin, in calcium oxalate crystallization in vitro. We studied a group of 10 male stone formers who had formed at least one...

Immunomodulatory properties of Tamm-Horsfall glycoprotein (THP) and uromodulin.

BACKGROUND Tamm-Horsfall glycoprotein (THP) and uromodulin are the most abundant glycoproteins in non-pregnant women's/men's and pregnant women's urine, respectively. However, the bioactivities of these glycoproteins are still unclear. OBJECTIVE To evaluate the immunomodulatory properties of THP and uromodulin on human peripheral blood mononuclear cells (PBMC) METHODS: THP and uromodulin isol...

Tamm-Horsfall protein or uromodulin: new ideas about an old molecule.

More than 50 years ago, Tamm and Horsfall isolated a mucoprotein from the human urine, and showed that the protein was able to interact and inhibit viral haemagglutination [1,2]. Of interest, the protein was found to be heavily glycosylated, containing up to 30% of its mass in carbohydrates [3]. It was then discovered that the Tamm–Horsfall protein (THP), as it was readily named, was the most a...

Mutant tamm-horsfall glycoprotein accumulation in endoplasmic reticulum induces apoptosis reversed by colchicine and sodium 4-phenylbutyrate.

As a consequence of uromodulin gene mutations, individuals develop precocious hyperuricemia, gout, and progressive renal failure. In vitro studies suggest that pathologic accumulation of uromodulin/Tamm-Horsfall glycoprotein (THP) occurs in the endoplasmic reticulum (ER), but the pathophysiology of renal damage is unclear. It was hypothesized that programmed cell death triggered by accumulation...

An Indirect Lectin Affinity Bioassay for Uromodulin

Uromodulin, also known as Tamm-Horsfall glycoprotein (THG), derived from human pregnancy urine, is a monomeric glycoprotein of 85 kDa. It is produced in the thick ascending limb (TAL) and early distal convoluted tubule (DCT) and is excreted in large amounts in urine. The immunosuppressive property of Uromodulin is based on its glycosylation pattern that is different THG obtained from males and ...
